Mechanism
Nocistatin is a natural peptide that blocks pain signaling by opposing the effects of nociceptin. It comes from the same precursor gene but has the opposite effect.
Technical detail
Nocistatin is derived from prepronociceptin (ppN/OFQ) and functionally antagonizes nociceptin/orphanin FQ-induced hyperalgesia and allodynia. It does not bind NOP receptors directly but instead modulates pain via distinct mechanisms possibly involving NMDA receptor signaling and inhibition of nociceptin release.
