Peptides for Anxiety — Which Ones to Consider and How

SELANK (tuftsin analog): The gold standard for peptide-based anxiety. Clinical trials in 60 humans showed anxiolytic effects comparable to phenazepam (a benzodiazepine), with effects lasting 1 week after stopping. Mechanism: modulates GABAergic system via GABA receptor interaction, plus minor BDNF upregulation. Key advantage: non-habit-forming, no sedation, mild nootropic co-effect. Dose: 250-1000 mcg intranasally per day (split into 2 doses), 10-20 day cycles. Works best for generalized anxiety, phobias, and somatoform disorders. N-Acetyl Selank Amidate is the more bioavailable modified form. SEMAX (ACTH 4-10 analog): Best for anxiety secondary to stress, burnout, or cognitive strain. Normalizes anxiety specifically when anxiety is elevated (no effect at baseline anxiety levels — a key safety feature). Mechanism: modulates monoamines (serotonin, dopamine, norepinephrine), BDNF, and neuroinflammatory pathways. Also enhances focus and executive function. Dose: 200-500 mcg intranasally per day, cycles of 2-4 weeks. N-Acetyl Semax Amidate is the modified high-potency form. OXYTOCIN: Best for social anxiety, trust deficits, and fear-of-others. Directly attenuates amygdala reactivity to social threats. Phase 3 trial (carbetocin, an oxytocin analog) showed significant reduction in anxiousness and distress in Prader-Willi syndrome. Mechanism: amygdala dampening via oxytocin receptors. Dose: 20-40 IU intranasally as needed. Effect duration 30-90 minutes. BPC-157: Indirect anxiolytic via gut-brain axis normalization. No direct human anxiety studies, but animal data shows stress reduction, GABAergic modulation, and restoration of dopamine system balance. Best suited when anxiety accompanies gut issues, chronic inflammation, or tissue injury recovery. SEMAGLUTIDE AND GLP-1 AGONISTS: 2024 animal data shows semaglutide reduces anxiety-like behaviors in T2D models via gut-brain axis (microbiota modulation, hippocampal neuroinflammation reduction, serotonin upregulation). This may explain why many users of semaglutide/tirzepatide report mood improvements.

💡 Tips

COMPARISON GUIDE: Selank = best for pure anxiety and phobias. Semax = best when anxiety co-occurs with brain fog or stress overload. Oxytocin = best for social anxiety specifically. BPC-157 = best when gut health or inflammation is contributing to anxiety. STACKING: Selank + Semax is a well-studied combination with synergistic effects (different mechanisms). Can be used on alternating days or combined in the same intranasal session. Selank + diazepam combination shows enhanced anxiolytic effect in stress conditions. RED FLAGS to discuss with a practitioner: If anxiety is severe, panic disorder is present, or suicidal ideation exists — these peptides are NOT replacements for clinical psychiatric care. They may be useful as adjuncts. FIRST-TIMER TIP: Start Selank at the low end (250 mcg) and assess response before increasing. Effects are often subtle — think less edge, better sleep, calmer baseline rather than dramatic sedation.
